Peptide Reconstitution: Laboratory Procedure
A standardized laboratory protocol for reconstituting lyophilized research peptides for in vitro use.
Purpose
To reconstitute a lyophilized research peptide into solution for in vitro laboratory experiments while preserving integrity and minimizing contamination.
Materials
- Lyophilized peptide vial
- Bacteriostatic water or specified buffer
- Sterile syringes and needles
- Alcohol wipes
Equipment
- Analytical balance (if weighing)
- Refrigerator (2–8°C)
- Vortex mixer (optional)
Procedure
- Allow the vial to equilibrate to room temperature before opening.
- Wipe the vial stopper with an alcohol wipe and allow to dry.
- Calculate the required solvent volume for the desired concentration.
- Draw the solvent into a sterile syringe.
- Inject the solvent slowly down the side of the vial (not directly onto the powder).
- Gently swirl (do not shake vigorously) until dissolved.
- Aliquot if needed and label with concentration and date.
Data Analysis
Record the final concentration, solvent used, and date. Verify by mass or volume as appropriate to your protocol.
Quality Control
Inspect the solution for clarity; particulates may indicate incomplete dissolution or contamination. Do not use compromised material.
Safety & Disposal
Wear gloves and eye protection. Dispose of sharps and unused material per institutional waste protocols.
