Overview

Purity testing verifies that a research compound meets its stated specification. For peptides, high-performance liquid chromatography (HPLC) is the primary quantitative method.

HPLC Analysis

Reverse-phase HPLC (RP-HPLC) separates peptide species by hydrophobicity. The target peptide and its impurities (truncated sequences, deamidation products, oxidation products) elute at different retention times, producing a chromatogram.

Purity is typically reported as the area percent of the main peak relative to the total integrated area at a specified detection wavelength (commonly 214 nm or 220 nm for peptides).

Mass Spectrometry

HPLC quantifies purity; liquid chromatography-mass spectrometry (LC-MS) confirms identity by measuring the molecular weight of the main peak. Together, HPLC + MS provide both purity and identity assurance.

Interpretation

  • A single dominant peak with ≥ 99% area suggests high purity.
  • Minor peaks indicate impurities; their identity can be investigated by MS.
  • Baseline resolution and symmetric peak shape indicate good chromatographic performance.

Acceptance Criteria

BlackBioLabs compounds are specified to a minimum purity of ≥ 99% by HPLC unless otherwise stated on the Certificate of Analysis.
